We downloaded and optimized the mature peptide sequence so that the eukaryote-derived protein may be suitable for expression in a prokaryotic expression system.

We optimized the sequence of tachypleus tridentatus clotting factor G alpha subunit, an eukaryotic protein, downloaded from GenBank, for more efficient protein expression in prokaryotic e.coli BL21 cells.We then inserted the optimized sequence into the multiple cloning site of pET-28a plasmid, which we downloaded from SnapGene.
